AM833 / Cagri: CagriSema Trial Evidence, Approval Status & Verified Vendor Prices
Cagrilintide (AM833) — Long-Acting Amylin Analogue
Last reviewed: 14 October 2026
Cagrilintide is Novo Nordisk's long-acting version of amylin, the hormone released alongside insulin that tells you you are full. On its own it produced about 11% weight loss in a phase 2 trial; paired with semaglutide as CagriSema it produced about 20% in phase 3, and that combination is under FDA review.
It activates amylin and calcitonin receptors in the brain to reduce appetite, and slows how fast food leaves the stomach. Because that is a different pathway from GLP-1, adding it to semaglutide gave more weight loss than semaglutide alone in the REDEFINE 1 trial.
It is the first amylin analogue to reach a phase 3 obesity programme, and the pairing with semaglutide is Novo's answer to tirzepatide and retatrutide. The research market picked it up in 2026 and it is now sold by most GLP-class vendors, often alongside blends.
Not approved anywhere; the combination is under review with a decision expected late 2026. Research-grade cagrilintide has no evidence of its own. Verify identity and the lot certificate at the laboratory, and do not compare a blend vial with a single-compound vial.
Cagrilintide (AM833) is a long-acting amylin analogue developed by Novo Nordisk. Amylin is the hormone co-secreted with insulin by pancreatic beta cells that slows gastric emptying and signals satiety through the area postrema and hypothalamus. Cagrilintide keeps that activity, adds activity at calcitonin receptors, and is engineered for once-weekly subcutaneous exposure through a fatty diacid side chain that binds albumin.
It is not approved anywhere on its own and has no regulatory filing as a single agent. Its route to market is the fixed-dose combination with semaglutide, CagriSema: Novo Nordisk filed the New Drug Application with the FDA on 18 December 2025 on the REDEFINE 1 and REDEFINE 2 results, a decision is expected in the last quarter of 2026, and in February 2026 the REDEFINE 4 head-to-head missed non-inferiority against tirzepatide. Status verified 14 October 2026.
On the research market cagrilintide is newer than the GLP-class compounds. Most vendors list it by name or as "Cagri"; blends with semaglutide (CagriSema-style) and with retatrutide are sold at a range of ratios and are not interchangeable with single-compound vials. The vendor table on this page compares single-compound vials only.
Cagrilintide is a 37-residue acylated peptide built on the human amylin backbone. Three proline substitutions (the same strategy used in pramlintide) prevent the fibrillation that makes native amylin unusable as a drug, calcitonin-derived residues in the mid-region broaden receptor engagement, and a C20 fatty diacid attached through a linker binds albumin to extend the half-life to roughly one week. The developer has not published the full sequence; vendor certificates identify it by mass and retention time against a reference standard.
Cagrilintide copies the satiety hormone amylin and lasts a week per dose: it acts on amylin and calcitonin receptors in the brainstem and hypothalamus, a satiety pathway separate from GLP-1, which is why it is being combined with semaglutide.
Cagrilintide is a dual amylin and calcitonin receptor agonist. Amylin receptors are complexes of the calcitonin receptor with receptor activity-modifying proteins (RAMP1–3); cagrilintide activates both the amylin receptor complexes and the calcitonin receptor itself, which is why it is described as a DACRA (dual amylin and calcitonin receptor agonist).
The weight effect is mainly central: amylin-receptor signalling in the area postrema and hypothalamus reduces food intake and increases satiety through a pathway distinct from GLP-1. That is the rationale for pairing it with semaglutide: the two act on separate satiety circuits, and in the REDEFINE 1 trial the combination produced more weight loss than semaglutide 2.4 mg alone at the same 68-week timepoint. Peripherally, amylin agonism slows gastric emptying and suppresses post-meal glucagon.
Activates calcitonin-receptor/RAMP complexes in the area postrema and hypothalamus, reducing food intake and increasing satiety.
Direct calcitonin-receptor activity in addition to amylin-receptor activity; the basis of the DACRA classification.
Slows gastric emptying and suppresses post-prandial glucagon, as native amylin does.
C20 fatty diacid side chain binds albumin, extending the half-life to about a week.
