Retatrutide is a triple agonist targeting GLP-1, GIP, and glucagon receptors. Phase 3 clinical trials show approximately 24.2% body weight reduction — the highest of any GLP-1 class compound in clinical development.
Phase 3 Clinical Trial Data
TRIUMPH-1 Phase 3 data: 24.2% mean body weight reduction at 48 weeks with 12 mg dose. This compares to 14.9% for semaglutide (STEP-1) and 20.9% for tirzepatide (SURMOUNT-1). Retatrutide has not yet received FDA approval as of 2026.
Titration Schedule
Research protocols mirror Phase 3 titration: 2 mg × 4 weeks → 4 mg × 4 weeks → 8 mg × 4 weeks → 12 mg maintenance. Slow titration is critical to minimize GI side effects.
Triple Receptor Mechanism
GLP-1 agonism: appetite suppression and glucose regulation. GIP agonism: enhanced insulin secretion and fat metabolism. Glucagon agonism: increased energy expenditure and hepatic fat reduction. The glucagon component distinguishes retatrutide from semaglutide and tirzepatide.
